1. The fastest would be a taxi. But it would cost you about $200-250 one-way.
2. You can take Interbus, it's a shuttle bus. You would go MA to San Jose and then SJ to Manzanillo. That would cost about $90 one-way.

Just throwing out another idea for a shuttle, Insterbus is a good shuttle company and i have also used anywhere costa rica's shuttle service as well. Their shuttle leaves at 7am from Manuel Antonio to Puerto Viejo (close to Manzanillo), around a 7.5 hrs. http://www.anywherecostarica.com/transpo...
I have also taken the bus and its not bad, look at it as a cultural experience and if $ is a concern it's a good option.

Why not rent an SUV at the airport or have a cab take you to one of the other nearby car rental places ? Get the smallest 4 wheel drive SUV available, they have 4 doors and are very reasonable and the rental agents will give you a map and any directions you want. Bueno Suerte !

What is the best and easiest way to get from the Pacific coast (Manuel Antonio) to the Caribbean coast (Manzanillo) I don't want to get the slow public bus back to San Jose and then another bus to the coast it would take 2 days.
